Development and Characterization of Solid Dispersion of Rasagilline Mesylate for Improvement of Dissolution Rate Using Hydrophilic Carriers

The aim of present study was to improve the solubility Rasagilline mesylate, an Atypical Antipsychotic agent which is BCS Class III drug and thus has very low hence poor bioavailability owing less absorption. Moreover mesylate a bitter taste. Thus in attempt made taste mask oral by formulating into solid dispersion then formulate dispersions Fast Dissolving Tablets. Solid Dispersions were formulated using various polymers like Plasdone K30, plasdone K90, Eudragit RS100, RL 100, Poloxamer 188 Soluplus different ratios viz., 1:1 1:2 solvent evaporation method evaluated for physicochemical parameters. On basis evaluation parameters Dispersion containing Drug : :: 1 2 optimized batch having Solubility 1.91 mg/ml, content 102.75 % CDR 98.43 at 35 mins. For Tablets, 32 factorial designs applied where SD12 Concentration Chitosan taken as dependent variable X1 X2, respectively wetting time (sec.) vitro disintegration independent variables. All formulation prepared applying experimental design showed more than 93 release 15 Batch F9 selected from data overlay plot, kinetics checkpoint batch. A stability carried out per ICH guidelines, no significant changes it can be concluded that Tablets provide rapid within short period time.
